As far as not having to hold the pad or note to play the whole sample, just turn the release all the way on the device you're using to play back your samples. As far as the looping, when you copy and paste it should loop perfectly AS LONG as you have the tempo right and you've chopped the sample right.
To be honest even though reason is pretty capable in sampling, it's far from the best at it. Partly because you cant really do any indepth audio editing, you can't timestretch it, and you can't record any audio. There's some other excellent programs that can handling sampling pretty easily, and can also rewire with reason so you can use them both at the same time, in sync.
And yes since your pads are sending the same midi notes as your actual keys, you should be able to play any instrument just fine (although I don't know why you would want to play a instrument using pads lol)
